Sarafloxacin ELISA

Sarafloxacin is a fluoroquinolone antibacterial drug. Like other (fluoro)quinolones, it acts by inhibiting the structure and function of DNA gyrase, a bacterial topoisomerase II which is an essential enzyme for DNA replication and transcription. Sarafloxacin has been proposed for use in the drinking water of poultry to treat bacterial infections caused by Escherichia coli and Salmonella spp, and in fish feed to treat diseases such as furunculosis, vibriosis and enteric redmouth.

The sarafloxacin ELISA is a competitive enzyme immunoassay for the screening and quantitative analysis of the antibiotic sarafloxacin in various matrices. The test is based on rabbit polyclonal antibodies against sarafloxacin.

EuroProxima has developed an ELISA for the screening of sarafloxacin residues in poultry and fish. In this ELISA polyclonal rabbit antibodies are used that were raised against protein bound sarafloxacin, whereas the conjugate consists of sarafloxacin labeled with horseradish peroxidase.

The polyclonal antibody only shows affinity for sarafloxacin (100% cross-reactivity) while difloxacin only shows 8% cross-reactivity. None of the other fluoroquinolones tested show affinity with this antiserum

The limit of detection (LOD) was determined for chicken tissue and salmonidae (muscle and skin). The LOD was found to be 4 ng/g (tissue samples) and 1 ng/g (salmonidae).
