Difloxacin ELISA

Difloxacin is a fluoroquinolone antibacterial drug. The compound has a broad antibacterial spectrum and is currently used in poultry, cattle and pigs for the treatment and control of difloxacin-sensitive infection. When used in the drinking water of poultry, difloxacin has a broad antibacterial spectrum with high bacterial activity against common avian pathogens. In both cattle and pigs, difloxacin is indicated for the treatment of respiratory diseases. For pigs it is also indicated for the treatment of Salmonellosis, piglet scours and enteritis/toxaemia.

The difloxacin ELISA is a competitive enzyme immunoassay for the screening and quantitative analysis of the antibiotic difloxacin in various matrices. The test is based on rabbit polyclonal antibodies against difloxacin.

EuroProxima has developed an ELISA for the screening of difloxacin residues in poultry and fish. In this ELISA polyclonal rabbit antibodies are used that were raised against protein bound difloxacin, whereas the conjugate consists of difloxacin labeled with horseradish peroxidase. The polyclonal antibody is rather specific for difloxacin and has no affinity for other fluoroquinolones tested.

The limit of detection (LOD) was determined for chicken, bovine and porcine tissue. The LOD was found to be 10, 8 and 8 ng/g, respectively.
